vaccine)\uff0c\u524d\u6b21\u5927\u6d41\u884c\u7684MERS-CoV\u8207SARS-CoV\u75ab\u82d7\u7814\u767c\uff0c\u4e5f\u6b77\u7d93\u5169\u5e74\u7814\u767c\u624d\u9032\u5165\u4e00\u671f\u81e8\u5e8a\u8a66\u9a57\uff0c\u800c\u5f9e\u5404\u5927\u85e5\u5ee0\u4e2d\u812b\u7a4e\u800c\u51fa\uff0c\u6253\u6557\u80dc\u80bd\u75ab\u82d7\u3001DNA\u75ab\u82d7\u3001\u817a\u75c5\u6bd2\u75ab\u82d7\u548c\u86cb\u767d\u8cea\u6b21\u55ae\u5143\u75ab\u82d7\uff0c\u5275\u4e0b\u6700\u5feb\u53d6\u5f97\u7dca\u6025\u6388\u6b0a\u7684\u75ab\u82d7\uff0c\u5206\u5225\u662f\u7531Pfizer-BioNTech\u8207Moderna\u7814\u767c\u7684\u5169\u6b3e\u53f2\u7121\u524d\u4f8b\u7684mRNA\u75ab\u82d7\uff0c\u53ea\u82b1\u4e8611\u500b\u6708\uff0c\u800c\u5176\u4e2d\u67098\u500b\u6708\u82b1\u5728\u81e8\u5e8a\u8a66\u9a57\u4e0a\u3002mRNA\u75ab\u82d7\u4e4b\u6240\u4ee5\u80fd\u6709\u5982\u6b64\u5feb\u901f\u7684\u7814\u767c\u6642\u7a0b(Moderna\u53ea\u82b1\u4e86\u5169\u5929\u5c31\u5efa\u7acb\u4e86mRNA\u5e8f\u5217\uff0c\u4e26\u572866\u5929\u5167\u5373\u9032\u5165\u7b2c\u4e00\u671f\u81e8\u5e8a\u8a66\u9a57)\uff0c\u9664\u4e86\u5927\u7b46\u7814\u7a76\u7d93\u8cbb\u7684\u6ce8\u5165\u8207\u7814\u767c\u5718\u968a\u7684\u52aa\u529b\u4e4b\u5916\uff0c\u4e5f\u662f\u57fa\u65bc\u591a\u5e74\u4f86\u5c0d\u65bc\u51a0\u72c0\u75c5\u6bd2\u3001siRNA\u905e\u9001\u8f09\u9ad4\u8207\u904e\u5f80mRNA\u4e4b\u7814\u767c\u7d93\u9a57\u4e0a\u3002<\/p>\n\n\n\n<p><strong>mRNA<\/strong><strong>\u8207\u975e\u75c5\u6bd2\u8f09\u9ad4<\/strong><br>SARS-CoV-2 mRNA\u75ab\u82d7\u7684\u539f\u7406\uff0c\u662f\u5c07\u53ef\u4ee5\u8868\u73fe\u75c5\u6bd2\u68d8\u86cb\u767d\u6210\u5206\u7684mRNA\u5e8f\u5217\u85c9\u7531\u975e\u75c5\u6bd2\u8102\u8cea\u5fae\u8f09\u9ad4\u905e\u9001\u5165\u4eba\u9ad4\u7d30\u80de\u4e2d\u7522\u751f\u68d8\u86cb\u767d\uff0c\u4e26\u85c9\u6b64\u523a\u6fc0\u514d\u75ab\u7cfb\u7d71\u7522\u751f\u5c08\u4e00\u6027\u514d\u75ab\u53cd\u61c9\u3002\u7136\u800c\u75ab\u82d7\u7684\u69cb\u6210\uff0c\u4e26\u4e0d\u662f\u7c21\u55ae\u7684\u5c07mRNA\u548c\u5fae\u8102\u7c92\u6df7\u5408\u5c31\u5b8c\u6210\u4e86\uff01mRNA\u4e4bUTR\u90e8\u5206\u9700\u7d93\u904e\u4fee\u6539\u4ee5\u907f\u514d\u88ab\u9175\u7d20\u964d\u89e3\uff0c\u85c9\u6b64\u5ef6\u9577\u514d\u75ab\u523a\u6fc0\u7684\u6642\u9593\uff1b\u70ba\u4e86\u589e\u52a0\u86cb\u767d\u8cea\u5408\u6210\u6548\u7387\uff0c\u800c\u9032\u884c\u5bc6\u78bc\u5b50\u7f6e\u63db\u4ee5\u589e\u52a0\u8f49\u8b6f\u901f\u7387\uff1b\u6b64\u5916\u91cd\u8981\u7684\u4e00\u9ede\u662f\uff0c\u672a\u7d93\u904e\u4fee\u98fe\u7684mRNA\u6838\u9178\uff0c\u6703\u88abpattern recognition receptors\u5982Toll-like receptor (TLR) 3\u3001TLR7\u8207TLR8\u8fa8\u8b58\uff0c\u88ab\u8aa4\u8a8d\u70ba\u75c5\u539f\u9ad4\u4e4bRNA\u9020\u6210\u8f49\u8b6f\u88ab\u6291\u5236\uff0c\u56e0\u6b64\u5e38\u7528\u7b56\u7565\u70ba\u5c07uridine\u7f6e\u63db\u70bapseudouridine\u6216\u5176\u4ed6\u6838\u82f7\u985e\u4f3c\u7269\u3002\u7d93\u904e\u8af8\u5982\u6b64\u985e\u7684\u6539\u52d5\uff0c\u624d\u80fd\u5920\u7a81\u7834mRNA\u75ab\u82d7\u4e4b\u74f6\u9838\uff0c\u9020\u5c31\u5982\u4eca\u770b\u4f3c\u7c21\u55ae\u53c8\u7d93\u6fdf\u7684mRNA\u7814\u767c\u6d41\u7a0b\u3002<br>\u7531\u65bcmRNA\u5e36\u6709\u5927\u91cf\u8ca0\u96fb\u8377\uff0c\u5176\u5206\u5b50\u91cf\u53c8\u4ecb\u65bc10~1000 kDa\uff0c\u4e0d\u53ef\u80fd\u7368\u81ea\u7a7f\u904e\u4e5f\u662f\u5e36\u8ca0\u96fb\u7684\u7d30\u80de\u819c\uff0c\u4e14\u88f8\u9732\u5728\u5916\u7684mRNA\u4ea6\u6703\u88ab\u514d\u75ab\u7cfb\u7d71\u8207\u6838\u9178\u5206\u89e3\u9176\u6e05\u9664\u3002\u56e0\u6b64\u9700\u8981\u5177\u6709\u9ad8\u5b89\u5168\u6027\u8207\u7a69\u5b9a\u6027\u7684\u8f09\u9ad4\u4fdd\u8b77\u4e26\u904b\u9001mRNA\u9032\u5165\u7d30\u80de\u3002\u76ee\u524d\u901a\u904e\u7684\u5169\u6b3eSARS-CoV-2 mRNA\u75ab\u82d7(Pfizer-BionTech\u7684BNT162b2\u8207Moderna\u7684mRNA-1273)\u90fd\u662f\u63a1\u7528\u8102\u8cea\u5948\u7c73\u7c92\u5b50\u4f5c\u70ba\u8f09\u9ad4\uff0c\u7531\u56db\u5927\u5143\u7d20\u7d44\u6210\uff1a\u53ef\u89e3\u96e2\u8102\u8cea(\u53d6\u4ee3\u6b63\u96fb\u8377\u8102\u8cea\uff0c\u8f03\u5b89\u5168\u4e14\u53ef\u5305\u8986mRNA\u4e26\u4fc3\u9032mRNA\u7531\u6eb6\u9176\u9ad4\u4e2d\u91cb\u653e\u5230\u7d30\u80de\u8cea)\u3001\u81bd\u56fa\u9187\u985e\u8102\u8cea(\u7a69\u5b9a\u5948\u7c73\u7c92\u5b50)\u3001\u8f14\u52a9\u78f7\u8102\u8cea(\u7dad\u6301\u8102\u8cea\u6d41\u52d5\u6027)\u3001\u8207\u5177PEG\u4fee\u98fe\u4e4b\u8102\u8cea(\u907f\u514d\u5de8\u566c\u7d30\u80de\u541e\u566c\u8207\u5ef6\u9577\u4f5c\u7528\u6642\u9593)\u3002\u57fa\u65bcFDA\u4e4b\u524d\u5df2\u901a\u904e\u4e4bsiRNA\u85e5\u7269Onpattro\u4ea6\u4ee5\u8102\u8cea\u5948\u7c73\u7c92\u5b50\u4f5c\u70ba\u8f09\u9ad4\uff0c\u5df2\u88ab\u8b49\u5be6\u4e4b\u5b89\u5168\u6027\u4ea6\u52a0\u901f\u4ee5\u6b64\u985e\u8f09\u9ad4\u4f5c\u70bamRNA\u75ab\u82d7\u4e4b\u6838\u51c6\u901f\u5ea6\u3002<\/p>\n\n\n\n<p><strong>\u5c0d\u61c9SARS-CoV-2\u4e4bmRNA\u75ab\u82d7<\/strong><br>\u622a\u81f32021\u5e748\u6708\uff0c\u5df2\u7d93\u6709185\u9805\u91dd\u5c0dSARS-CoV-2\u7684\u75ab\u82d7\u5728\u9032\u884c\u81e8\u5e8a\u524d\u8a66\u9a57\uff0c\u4e5f\u6709102\u500b\u54c1\u9805\u6b63\u5728\u9032\u884c\u81e8\u5e8a\u8a66\u9a57\uff0c\u5176\u4e2d\u670919\u500b\u662fmRNA\u75ab\u82d7\u3002BNT162b2\u8207mRNA-1273\u5c55\u73fe\u4e86\u5927\u65bc90%\u7684\u4fdd\u8b77\u529b\u6578\u64da\uff0c\u4e26\u5177\u6709\u76f8\u5c0d\u8f15\u5fae\u7684\u85e5\u7269\u526f\u4f5c\u7528(\u6ce8\u5c04\u8655\u75bc\u75db\u8207\u767c\u71d2)\uff0c\u88ab\u8996\u70ba\u662f\u5c0d\u5927\u773e\u4f86\u8aaa\u8f03\u5b89\u5168\u7684\u75ab\u82d7\u9078\u64c7\u3002\u7136\u800c\u5176\u904b\u8f38\u53ca\u4fdd\u5b58\uff0c\u9700\u914d\u5408\u5b8c\u6574\u7684\u51b7\u934a\u7cfb\u7d71\uff0cmRNA-1273\u57284-8\u00b0C\u51b7\u85cf\u53ef\u4fdd\u5b58\u4e00\u500b\u6708\uff0c\u5728\u5ba4\u6eab\u53ea\u80fd\u4fdd\u5b5812\u5c0f\u6642\uff0c\u800cBNT162b2\u5247\u9808\u4fdd\u5b58\u65bc-60\u00b0C`\uff0c\u5927\u5e45\u9650\u5236\u5176\u5728\u8f03\u843d\u5f8c\u570b\u5bb6\u7684\u65bd\u6253\u3002\u70ba\u89e3\u6c7a\u6eab\u5ea6\u7684\u9700\u6c42\uff0c\u6709\u6578\u500b\u5c0d\u6eab\u5ea6\u8981\u6c42\u8f03\u5bec\u9b06\u7684\u75ab\u82d7\u5019\u9078\u8005\u6b63\u5728\u9032\u884c\u7814\u767c\u8207\u81e8\u5e8a\u8a66\u9a57\uff0c\u5982CureVac\u7684CVnCoV\u53ef\u57285\u00b0C\u4fdd\u5b583\u500b\u6708\u3001Walvax Biotechnology\u7684ARCoV\u53ef\u572825\u00b0C\u4fdd\u5b58\u4e00\u9031\u7b49\uff0c\u7686\u662f\u4ee4\u4eba\u671f\u5f85\u7684\u7522\u54c1\u3002\u53e6\u5916\uff0c\u53ef\u81ea\u6211\u8907\u88fd\u4e4bmRNA\u75ab\u82d7\u4e5f\u662f\u5177\u6f5b\u529b\u7684\u75ab\u82d7\u5019\u9078\u3001\u900f\u904e\u53ef\u5728\u7d30\u80de\u5167\u8907\u88fd\u4e4b\u6a5f\u8f49\uff0c\u80fd\u5920\u6e1b\u5c11\u6700\u521d\u65bd\u6253\u5291\u91cf\u4ee5\u964d\u4f4e\u526f\u4f5c\u7528\u3002\u502b\u6566\u5e1d\u570b\u5927\u5b78\u8207Acuitas Therapeutics\u5171\u540c\u7814\u767c\u4e4bLNP- nCoVsaRNA\u5df2\u5728\u9032\u884c\u81e8\u5e8a\u8a66\u9a57\u3002<\/p>\n\n\n\n<p><strong>COVID-19<\/strong><strong>\u4e4b\u5916\u7684\u611f\u67d3\u75c7<\/strong><br>\u65e9\u5728\u91dd\u5c0dSARS-CoV-2\u4e4b\u524d\uff0c\u8af8\u591a\u91dd\u5c0d\u4e0d\u8ad6\u662f\u764c\u75c7\u6216\u50b3\u67d3\u6027\u75be\u75c5\u7684mRNA\u75ab\u82d7\u958b\u767c\u5c31\u5df2\u7d93\u5728\u9032\u884c\uff0c\u5982\u6d41\u611f\u75c5\u6bd2(influenza virus)\u3001\u8332\u5361\u75c5\u6bd2(Zika virus)\u3001\u4eba\u985e\u514d\u75ab\u7f3a\u4e4f\u75c5\u6bd2(HIV)\u3001\u547c\u5438\u9053\u878d\u5408\u75c5\u6bd2(RSV)\u3001\u4f0a\u6ce2\u62c9\u75c5\u6bd2(Ebola virus)\u3001\u72c2\u72ac\u75c5\u6bd2(Rabies virus)\u3001\u8207\u7627\u539f\u87f2(malaria parasite)\u7b49\uff0c\u4f46\u622a\u81f32019\u5e74\u5e95\uff0c\u91dd\u5c0d\u4e0a\u8ff0\u50b3\u67d3\u75c5\u768415\u500bmRNA\u75ab\u82d7\u7686\u672a\u9032\u5165\u7b2c\u4e09\u671f\u81e8\u5e8a\u8a66\u9a57\u3002\u7136\u800c\u6709\u8457mRNA\u75ab\u82d7\u5728SARS-CoV-2\u7684\u6210\u529f\u7d93\u9a57\uff0c\u8207\u5c0d\u61c9\u5404\u75be\u75c5\u4e4b\u5b8c\u6574\u81e8\u5e8a\u524d\u8cc7\u6599\uff0c\u61c9\u53ef\u52a0\u901fmRNA\u75ab\u82d7\u4e4b\u81e8\u5e8a\u8a66\u9a57\u8207\u61c9\u7528\u3002<\/p>\n\n\n\n<p><strong>\u76ee\u524dmRNA\u75ab\u82d7\u4e4b\u8003\u91cf<\/strong><br>\u75ab\u82d7\u4fdd\u8b77\u529b\u6301\u7e8c\u6642\u9593\u4e00\u76f4\u662f\u73fe\u4eca\u5c0d\u5404SARS-CoV-2\u75ab\u82d7\u7684\u6700\u5927\u8003\u9a57\u4e4b\u4e00\u3002\u514d\u75ab\u529b\u7684\u5f62\u6210\uff0c\u662f\u900f\u904emRNA\u8868\u73fe\u51fa\u6297\u539f\u86cb\u767d\u5f8c\uff0c\u88ab\u6297\u539f\u5448\u73fe\u7d30\u80de\u651c\u5e36\u81f3\u6dcb\u5df4\u7d50\uff0c\u8207\u5176\u4e2d\u7684B\u7d30\u80de\u548c\u6ffe\u6ce1\u8f14\u52a9T\u7d30\u80de(follicular helper T cell, T<sub>FH<\/sub>)\u5f62\u6210\u751f\u767c\u4e2d\u5fc3\uff08germinal center\uff09\uff0c\u9020\u6210\u6297\u539f\u5c08\u4e00\u6027B\u7d30\u80de\u5927\u91cf\u589e\u751f\u6d3b\u5316\uff0c\u4ee5\u88fd\u9020\u6297\u9ad4\u3002\u7136\u800c\u6839\u64da\u771f\u5be6\u4e16\u754c\u6578\u64da\u8207\u81e8\u5e8a\u8cc7\u6599\uff0c\u6297\u539f\u53ca\u8102\u8cea\u8f09\u9ad4\u7a2e\u985e\u7686\u6703\u5f71\u97ff\u6297\u9ad4\u53cd\u61c9\u6301\u7e8c\u6642\u9593\uff0c\u56e0\u6b64\u4ecd\u9700\u8981\u66f4\u9032\u4e00\u6b65\u7684\u7814\u7a76\u8207\u9577\u6642\u9593\u76e3\u6e2c\u624d\u53ef\u80fd\u91d0\u6e05\u3002<br>\u6b64\u5916\uff0c\u75c5\u6bd2\u7684\u5feb\u901f\u8b8a\u7570\uff0c\u65bd\u6253\u7684\u75ab\u82d7\u80fd\u5426\u63d0\u4f9b\u8db3\u5920\u4fdd\u8b77\u529b\uff0c\u4e5f\u662f\u5927\u90e8\u5206\u4eba\u64d4\u5fc3\u7684\u91cd\u8981\u8b70\u984c\uff0c\u5982HIV\u75ab\u82d7\u7684\u5931\u6557\uff0c\u90e8\u5206\u539f\u56e0\u4f86\u81ea\u65bc\u592a\u591a\u4e0d\u540c\u7684HIV\u8b8a\u7570\u682a\u3001\u6d41\u611f\u75ab\u82d7\u4e5f\u662f\u5fc5\u9808\u6bcf\u5e74\u66f4\u65b0\uff0c\u56e0\u6b64\u6311\u9078\u75c5\u6bd2\u4e4b\u76f8\u5c0d\u4fdd\u5b88\u4e4b\u86cb\u767d\u5e8f\u5217\u9032\u884c\u75ab\u82d7\u958b\u767c\uff0c\u662f\u4e00\u500b\u91cd\u8981\u7684\u8ab2\u984c\u3002SARS-CoV-2\u4e5f\u662f\u4e00\u500b\u8b8a\u7570\u901f\u5ea6\u5feb\u901f\u7684\u75c5\u6bd2\uff0c\u56e0\u6b64\u5982Moderna\u4ea6\u6b63\u5728\u7814\u767c\u91dd\u5c0d\u5f8c\u7e8c\u8b8a\u7570\u682a\u7684mRNA\u75ab\u82d7\u4f5c\u70ba\u88dc\u5145\u5291\u3002\u7136\u800c\u96a8\u8457\u86cb\u767d\u8cea\u7d50\u69cb\u5b78\u7684\u7cbe\u9032\uff0c\u767c\u5c55\u51fa\u91dd\u5c0d\u5404\u51a0\u72c0\u75c5\u6bd2\u4fdd\u5b88\u90e8\u4f4d\u7684\u75ab\u82d7\u4ecd\u662f\u5099\u53d7\u671f\u5f85\u3002<br>\u5c31\u5b89\u5168\u6027\u4f86\u8aaa\uff0c\u96d6\u7136mRNA\u75ab\u82d7\u5df2\u76f8\u5c0d\u5176\u4ed6SARS-CoV-2\u75ab\u82d7\u6709\u8457\u8f03\u8f15\u5fae\u7684\u4e0d\u826f\u53cd\u61c9\uff0c\u4ecd\u6709\u76f8\u5c0d\u65bc\u50b3\u7d71\u75ab\u82d7\u4f86\u8aaa\u8f03\u9ad8\u6bd4\u4f8b\u7684\u904e\u654f\u6027\u4f11\u514b\u3002\u5176\u4e2d\u4e00\u500b\u8f03\u53ef\u80fd\u7684\u5047\u8aaa\u662f\uff0c\u904e\u654f\u53cd\u61c9\u4f86\u81ea\u65bc\u8102\u8cea\u5948\u7c73\u7c92\u5b50\u7684PEG\u5206\u5b50\uff0c\u800c\u6700\u8fd1\u7f8e\u570bCDC\u4e5f\u767c\u5e03\u4e86\u4e00\u689d\u5efa\u8b70\uff0c\u4e0d\u61c9\u5c0dPfizer-BioNTech\u6216Moderna\u75ab\u82d7\u7684\u4efb\u4f55\u6210\u5206\u6709\u904e\u654f\u53cd\u61c9\u53f2\u7684\u4eba\u63a5\u7a2e mRNA\u75ab\u82d7\u3002\u56e0\u6b64\uff0c\u91dd\u5c0dmRNA\u8f09\u9ad4\u4e4b\u958b\u767c\uff0c\u61c9\u53ef\u589e\u52a0\u5176\u5b89\u5168\u6027\u3002<\/p>\n\n\n\n<p><strong>\u7d50\u8a9e\u8207\u5c55\u671b<\/strong><br>\u4ee5\u5f80\u7684\u89c0\u5ff5\uff0c\u8a8d\u70ba\u57fa\u65bc\u7a69\u5b9a\u6027\u8207\u4f5c\u7528\u539f\u7406\uff0cRNA\u662f\u6975\u4e0d\u53ef\u80fd\u5ee3\u6cdb\u4f5c\u70ba\u85e5\u7269\u65bd\u4e88\u7684\u3002\u7136\u800c\u7d93\u7531\u9019\u6b21\u7684COVID-19\u5927\u6d41\u884c\uff0c\u6a6b\u8de8\u751f\u7269\u5316\u5b78\u3001\u5316\u5b78\u3001\u751f\u7269\u8cc7\u8a0a\u3001\u85e5\u7269\u50b3\u8f38\u7b49\u9818\u57df\u5efa\u7acb\u7684mRNA\u75ab\u82d7\u5e73\u53f0\uff0c\u53ef\u671b\u958b\u555f\u65b0\u7684\u4e00\u6ce2\u75ab\u82d7\u7814\u767c\u6280\u8853\uff0c\u8207\u91dd\u5c0d\u4e0d\u8ad6mRNA\u75ab\u82d7\u5404\u7d30\u90e8\u74b0\u7bc0\u4e4b\u6539\u826f\u3002\u9664\u50b3\u67d3\u6027\u75be\u75c5\u4e4b\u5916\uff0c\u4ea6\u53ef\u80fd\u63a8\u5ee3\u81f3\u764c\u75c7\u75ab\u82d7\u4e4b\u7814\u767c\uff0c\u5c0d\u91ab\u5b78\u9818\u57df\u6975\u6709\u8ca2\u737b\u3002<br><br><strong>\u53c3\u8003\u8cc7\u6599\uff1a<\/strong><br>1.&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;
